Responsabilities

High Z metallic nanoparticles (NP) as radiosensitizers have been described as a candidate of choice for combined modalities in the last decade. New findings have highlighted mechanisms promoted by NPs related to reactive oxygen species (ROS) production through thioredoxine reductase (TrxR) inhibition and mitonchondria dysfunction. This study proposes to design a novel type of gold nanoparticle conjugated to an antibody targeting M2 macrophages (GNP) in a pancreatic tumor model, in vitro and in vivo. The use of GNPs in combination with proton radiation, which has previously been shown to favor a mixed M1/M2 phenotype, could lead to a full phenotype change to M1 macrophages. In the context of the emerging field of protontherapy, with two new centres opening soon in Belgium, it is of crucial importance to understand the impact of concomitant use of GNPs and charged particles on immune cells. The work will be carried out in the LARN laboratory at the University of Namur.

The applicant should:

• Develop the synthesis of gold nanoparticle and conjugation to antibody,

• Be able to work with animals and develop tumor models,

• Interact with academic partners for the development of antibodies and test for specificity in vitro and in vivo,

• Irradiate samples using the particle accelerator of UNamur and X-ray irradiator, and outside Belgium through privileged partnerships.

Profile

Applicant should have a PhD degree in biology, biochemistry or pharmacy. The applicant should be in mobility conditions:

having spent 12 months or more outside Belgium in the past 24 months.

• Certificate in animal science (authorized to conduct experiments)

• Analytical and synthetical mind

• Adaptation and organization

• Fluent in English
